Fluorescence Lifetime Measurement Apparatus

The sample is excited with SHG laser pulses (wavelength of pulses ~400 nm, fwhm ~ 100 fs of pulses). Fluorescence from the sample is dispersed with a monochromator and measured using a streak scope wavelength range from 400 to 800 nm, time range from 2 ps (2 × 10-12 s) to 1ms (1 × 10-6 s)). The fluorescence signals will be averaged using PC. Analysis of fluorescence decay (the time dependence of fluorescence intensity) provides the fluorescence lifetime (the period during which the fluorescence intensity decays to 1/e), which is 1.4 times the half-life value. Furthermore, the time-dependent fluorescence spectrum can be measured to obtain a three-dimensional contour map of the time- and wavelength-dependent fluorescence intensity. In the case of non-single exponential fluorescence decay, kinetic parameters can also be obtained from the analysis. For example, the energy transfer rate from the fluorophore to the energy acceptor can be obtained.
